RFC-destination and site for R/3 backend exists

Perform this step by selecting the option 'Enable Initial Load from R/3 backend'. Select the appropriate object for which the initial load should be enabled.

You can check the new entry for consumer by calling sm30 for table CRMCONSUM in your R/3 backend system. There must be an entry now with Consumer = CRM30.
